MY'STORY

The MOVE Fire

This is a personal recollection on the Move fire on May 13, 1985 Philadelphia police fired thousands of rounds at the MOVE house, city officials approved dropping an explosive device on the roof, the resulting fire was allowed to burn, 11 people—including five children—died, and 61 homes were destroyed. Philadelphia City Council later called it a “brutal attack carried out by the City of Philadelphia on its own citizens” and acknowledged that no individual faced criminal consequences for the bombing. One timeline correction worth preserving for the BHP record: the major previous MOVE-police confrontation was August 8, 1978, about seven years before the bombing, not a year or two earlier. Officer James Ramp was killed, other police and firefighters were wounded, nine MOVE members were later convicted, and television cameras recorded police beating Delbert Africa during his arrest. The 1985 MOVE Commission later specifically criticized city planners for failing to adequately use lessons from that 1978 confrontation. And that actually strengthens the point you’re making: 1985 did not happen without precedent or institutional memory. There had already been a deadly confrontation with MOVE, years of conflict, negotiations and police involvement before Osage Avenue.

Reuptake modulator

A reuptake modulator, or transporter modulator, is a type of drug which modulates the reuptake of one or more neurotransmitters via their respective neurotransmitter transporters.[1] Examples of reuptake modulators include reuptake inhibitors (transporter blockers)[2] and reuptake enhancers.[3]

MTHPI

MTHPI, also known as 3-(1-methyl-1,2,3,6-tetrahydropyrid-4-yl)indole (3-MTPI), is a serotonin receptor modulator of the tetrahydropyridinylindole group related to the psychedelic drug dimethyltryptamine (DMT). Though it is not technically a tryptamine itself, MTHPI can be regarded as a constrained analogue of DMT. The drug shows affinity for the serotonin 5-HT1, 5-HT1A, and 5-HT2A receptors (Ki = 217 nM, 144 nM, and 71 nM, respectively). Its affinity for the serotonin 5-HT2A receptor was 8-fold higher than that of DMT. Besides serotonin receptors, MTHPI also shows affinity for the serotonin transporter (SERT) (IC50Tooltip half-maximal inhibitory concentration = 690 nM). The chemical synthesis of MTHPI has been described. Derivatives of MTHPI such as 5-MeO-MTHPI, 2-Me-MTHPI, and 5-MeO-2-Me-MTHPI have also been described. 5-MeO-MTHPI showed 11-fold lower affinity for the serotonin 5-HT2A receptor and 6-fold higher affinity for the serotonin 5-HT1A receptor compared to MTHPI. Conversely, the 2-methylated analogues showed markedly reduced affinities for the assessed serotonin receptors. Various other derivatives of MTHPI, acting as monoamine transporter modulators, have been described as well. MTHPI was first described in the scientific literature by Ethan Will Taylor and colleagues by 1987.

TACT411

TACT411 is a monoamine transporter modulator and serotonin 5-HT1B receptor modulator which is under development for the treatment of alcohol use disorder, anorexia, and binge-eating disorder. It is being developed by Tactogen. As of the first quarter of 2025, the drug is in the preclinical research stage of development. The exact chemical structure of TACT411 does not yet appear to have been disclosed, but it may be a substituted benzofuran derivative.

1Z2MAP1O

1Z2MAP1O, also known as 1-(indolizin-3-yl)-2-(methylamino)propan-1-one, is a monoamine releasing agent and serotonin receptor modulator of the indolizinylethylamine family. It is an analogue of BK-NM-AMT (β-keto-N-methyl-α-methyltryptamine) in which the indole ring has been replaced with an indolizine ring. 1Z2MAP1O has been found to be a serotonin and dopamine releasing agent as well as a weak serotonin 5-HT1B receptor agonist and serotonin 5-HT2B receptor antagonist. Conversely, its activity as a norepinephrine releasing agent was not reported, and it was inactive at the remaining sites of the 47 screened targets. The drug's EC50Tooltip half-maximal effective concentration values for induction of monoamine release were 109 nM for serotonin and 227 nM for dopamine in Chinese hamster ovary (CHO) cells expressing the human monoamine transporters (MATs). The dopamine/serotonin ratio was 0.48. Its EC50 at the serotonin 5-HT1B receptor was 1,130 nM, whereas its IC50Tooltip half-maximal inhibitory concentration values were 2,140 nM at the serotonin 5-HT2B receptor, 2,410 nM at the dopamine transporter (DAT), and 5,690 nM at the norepinephrine transporter (NET). 1Z2MAP1O's inhibition of the serotonin transporter (SERT) was not reported. 1Z2MAP1O was patented by Matthew Baggott of Tactogen in 2023.
